The Influence of 5-Aminolevulinic Acid on Protochlorophyllide and Protochlorophyll Accumulation in Dark-Grown Scenedesmus
نویسندگان
چکیده
The interm ediate o f chlorophyll b iosynthesis, 5-am inolevulinic acid (A L A ), is a necessary prerequisite for the form ation o f protochlorophyllide (PChlide) and protochlorophyll (PChl) in the dark. The application o f A L A to a dark-grown culture o f the pigm ent m utant C-2 A' o f Scenedesmus obliquus increased the am ount o f PChlide 30-fold and the am ount o f PChl about 10-fold. The rates o f A L A -dependent form ation o f PChlide and PChl reach their m axim um values at different concentrations o f added A L A . Sim ilarly, the kinetics o f PChlide and PChl form ation in cells incubated with A L A are different. Cells o f Scenedesmus m utant C-2 A' incu bated with various concentrations o f A L A for different periods provide a good tool for future studies differentiating between PChlide and PChl m etabolism . — The incorporation o f Chi de riving from either PChl or PChlide into different pigm ent protein com plexes is discussed.

The Absence of Protochlorophyll(ide) Accumulation in Algae with Inhibited Chlorophyll Synthesis.
Golenkinia, Chlorella protothecoides, and mutant C-2A' of Scenedesmus were grown in darkness and on media in which chlorophyll synthesis is reduced significantly. The pigments were analyzed by spectrophotometry or by paper chromatography and compared with similar extracts from light-grown algae and dark-grown beans. No protochlorophyll(ide) was present in the dark-grown algae indicating that ch...

The enzymic nature of the protein moiety of protochlorophyll(ide) holochrome was studied by following the fate of the [(14)C]protochlorophyll(ide) formed when dark-grown barley (Hordeum vulgare) or bean (Phaseolus vulgaris) leaves are incubated in the dark with 3 mm 4-delta-[(14)C]aminolevulinic acid. It was found that: [List: see text]Since turnover of protochlorophyll(ide) was not observed, t...

A study of greening in cucumber (Cucumis sativus L.) cotyledons grown under a light (14-hour) dark (10-hour) photoperiodic regime was undertaken. The pools of protoporphyrin IX, Mg-protoporphyrin IX monoester, protochlorophyllide, and protochlorophyllide ester were determined spectrofluorometrically. Chlorophyll a and b were monitored spectrophotometrically. Pigments were extracted during the 3...

متن کاملProtochlorophyll biosynthesis in a cell-free system from higher plants.
A cell free system prepared from etiolated cucumber (Cucumis sativus, L) in tris-sucrose buffer is able to incorporate delta-aminolevulinic acid-4- (14)C into the two components of protochlorophyll: protochlorophyllide and protochlorophyllide ester. The activity is associated with the etioplasts. Optimal incorporation is obtained at pH 7.7. For the formation of protochlorphyllide ester, oxygen,...
